Isolasi Protein Dan Analisis Asam Amino Konsentrat Protein Daun Kaliandra Sebagai Upaya Penyediaan Suplemen Pakan Ternak: Kualitas Asam Amino Konsentrat Protein Hasil Ekstraksi Daun Kaliandra
Abstract
The Research aims to determine the effect of extraction using various combination of pH acetat buffer solution pH 4,0; 4,6 and 5,2 to amino acid content of Caliandra Leaf Protein Concentrate (CLPC). Completely Randomized Design (CRD) with 3 treatments and 5 replications. The Caliandra Leaf Meal (CLM) (50 gram) was blended in acetat buffer solution (200 ml) pH 4,0; 4,6 and 5,2 for 15 menit and centrifuged to separate the supernatant from extracted CLMandprecipitated with buffer mix phospat (citric acid and Na2HPO4) until isoelectric point. The treatments were tested consisted of extracted with acetat buffer solution pH 4,0; 4,6 and 5,2. The parameters measured; isoelectric point at precipited, total non essential amino acid (TNEAA), total essential amino acid (TEAA) and Essential Amino Acid Index (EAAI).Amino acids content analysis by high performance liquid chromatography (HPLC). The results of analysis by HPLC showed that the content of total amino acids and essential amino acids index of Caliandra Leaf Protein Concentrate (CLPC) decreased with increasing pH buffer solution, except for the total content of the essential amino acid buffer solution pH 4.0 is lower than pH 4.6. CLPC contains all of amino acids except asparagine, glutamine, and tryptophan.
